Thrombospondin is a protein from platelet a-granules. It is made up of three identical subunits bound by interchain disulfides. It is secreted at sites of platelet activation and aggregation and is involved in the processes of chemotaxis, adhesion, proliferation and differentiation of leukocytes, fibroblasts, smooth muscle and endothelial cells.

ApplicationsSuitable for use in Electron Microscopy, Immunoprecipitation, and Western Blot. Not suitable for Immunohistochemistry.
Other applications not tested.
Recommended DilutionImmunoprecipitation (Native verified): 2ug/mg protein lysate. Use Protein G.
Western Blot: 1-2ug/ml for 2hrs at RT. Detects ~450kD band (non-reducing) or 170-180kD band (reducing)
Optimal dilution to be determined by the researcher.

Storage and StabilityMay be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

ImmunogenReduced and alkylated puri ed human TSP (fully denatured) from the supernatant of thrombin-activated platelets. Epitope: Calcium-binding domain of TSP (C-terminal 50kD piece of the 120kD fragment).
SpecificityRecognizes human TSP1. Reacts with 140kD and 120kD fragments in digest of Ca2+-replete TSP which are absent from digest in EDTA. Its binding to TSP is enhanced by EDTA i.e. at low [Ca2+]. Does not crossreact with fibronectin, fibrinogen, TSP-2 and von Willebrand factor. Species Crossreactivity: mouse, rat, bovine, porcine, equine, canine and sheep.
