| Thyroid hormone receptors (TRs) belong to a large superfamily of DNA-binding proteins, which includes the steroid hormone, vitamin D, and retinoic acid receptors. Members of this family share a similar domain organization that features a central DNA-binding domain containing two zinc fingers and carboxy-terminal ligand-binding domain. Three TR isoforms have been identified in the rat. THR, beta-1 and THR, beta-2 are encoded on the same gene and differ only at the amino-terminal region due to alternative splicing and promoter selection. THR, alpha-1, which is encoded on a separate gene, is homologous to the THR, beta in both the DNA and ligand-binding domains. Interestingly, an alternative splice product of THR, alpha-1, c-erbA, alpha-2, has a different carboxy-terminal region and does not bind thyroid hormone. |
