| Toll-like receptors (TLRs) are evolutionarily conserved pattern-recognition molecules resembling the toll proteins that mediate antimicrobial responses in Drosophila. These proteins recognize different microbial products during infection and serve as an important link between the innate |
|
| and adaptive immune responses (1,2). The TLRs act through adaptor molecules such as MyD88 and TIRAP to activate various kinases and transcription factors (3) so the organism can respond to potential infection. TLR1 is expressed at the cell surface as a heterodimer with either TLR2 or TLR6. These dimers recognize GPI-linked proteins and di- and triacyl lipoproteins. TLR4 and TLR5 are also expressed at the cell surface and respond to LPS and bacterial flagellin, respectively. TLRs 3, 7, 8 and 9 are localized in intracellular acidic compartments such as the phagolysosome (4) and recognize viral RNA and CpG DNA sequences. TLR10 is homologous to TLR1 and TLR6 and highly expressed in lymphoid tissues, although it exists only as a pseudogene in mice (5). |
