| TLR4 is an 839aa member of the Toll-like receptor (TLR) family of transmembrane proteins comprised of multiple copies of leucine rich repeats in the extracellular domain and a TIR domain. It is a widely studied, phylogenetically conserved, member playing a fundamental role in pathogen recognition and activation of innate immunity. Like its counterpart in Drosophila, TLR4 signals through adaptor molecules and mediates the production of cytokines necessary for the development of effective immunity. TLR4 belongs to the multi-protein complex of lipopolysaccharide (LPS) receptor, containing CD14, LY96 and TLR4, and is involved in signal transduction events induced by lipopolysaccharide (LPS) found in most gram-negative bacteria. This receptor is most abundantly expressed in placenta, spleen and in myelomonocytic subpopulation of the leukocytes.
