Topoisomerase II alpha is one of the isoforms of the mammalian Topoisomerase II which has been implicated in many aspects of DNA metabolism, including transcription, replication, recombination, and segregation. It is preferentially expressed in proliferating cells and has been found to increase during S phase, peak at G2-M, and be diminished during G1. The gene encoding the human protein has been mapped to chromosome 17q21-22. It is an essential nuclear enzyme for chromosome segregation during mitosis. Mutations in the protein associated with drug resistance show clustering in exons 12, 13, 19-21 and 34-35. The pathological role of the enzyme has been identified the ataxia-telangiectasia.
