| The TRAF (tumor necrosis factor receptor-associated factor) family of proteins associate with and transduce signals from members of the tumor necrosis factor receptor (TNFR) superfamily. TANK:I-TRAF (TRAF family member associated NF-kB activator:TRAF-interacting protein) that was first defined as a novel TRAF-interacting protein that regulates TRAF-mediated signal transduction (reviewed in Bonif, 2006). TANK:I-TRAF is thought to inhibit TRAF function by sequestering the TRAFs in the cytoplasm, thereby blocking their recruitment to the TNFR. TANK:I-TRAF has also been shown to bind to other proteins in the NF-kB signaling pathway, including NEMO. However, the role and different functions of TANK:I-TRAF in NF-kB signaling pathways remains to be fully elucidated.
