| T2BP, a novel 184aa protein, is human homolog of mouse Tifa. It consists of a central phosphopeptide-binding FHA domain and a C-terminal TRAF6-binding motif. T2BP mediates multiple protein-protein interactions and plays a significant role in innate immunity, adaptive immunity and inflammation. It interacts with TRAF2 and is involved in TNF-mediated signaling. It induces oligomerization and polyubiquitination of TRAF6 leading to activation of TAK1 and IKK in a proteasome-independent mechanism and mediates NF-kappaB and JNK activation.
