Clone Host Grade Applications
Monoclonal Mouse Affinity Purified E B
Troponin, working in conjunction with tropomyosin, functions as a molecular switch, regulating muscle contraction in response to changes in the intracellular Ca2+ concentration. Troponin consists of three subunits, the Ca2+-binding subunit troponin C (TnC), the tropomyosin-binding subunit troponin T (TnT), and the inhibitory subunit troponin I (TnI). Troponin I binds to actin in thin myofilaments to hold the actin-tropomyosin complex in place, because of which myosin cannot bind to actin in relaxed muscle. When Calcium binds to the Troponin C it causes certain confomational changes which leads to dislocation of troponin I. The letter I is given due to its inhibitory character.
Catalog #T8665-16R
The tissue specific subtypes areSlow-twitch skeletal muscle isoform troponin I, TNNI1 (1q31.3, 191042)
Fast-twitch skeletal muscle isoform troponin I, TNNI2 (11p15.5, 191043)
Cardiac troponin I, TNNI3 (19q13.4, 191044)
ApplicationsSuitable for use in ELISA and Western Blot. Other applications not tested.
Recommended DilutionOptimal dilutions to be determined by the researcher.
Matched PairSuitable for use as the detection or conjugate (label) antibody.
Matched to T8665-16E as the capture or solid phase antibody.
Also suitable as the detection or conjugate (label) antibody with T8665-19B. Matched to T8665-16E and T8665-16H as the capture or solid phase antibodies.
HybridomaHybridization of Sp 2/0 myeloma cells with spleen cells from Balb/c mice.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

ImmunogenFree human Cardiac Troponin I
SpecificityRecognizes an epitope located between aa169-178 of cTnI. Reacts equally with free cardiac troponin I (cTnl) and cTnI forming complexes with other troponin components (in the presence of 5mM EDTA). Not affected by heparin, phosphorylation and oxidation. No crossreactivity with skeletal muscle troponin I.
