| Troponin, working in conjunction with tropomyosin, functions as a molecular switch, regulating muscle contraction in response to changes in the intracellular Ca2+ concentration. Troponin consists of three subunits, the Ca2+-binding subunit troponin C (TnC), the tropomyosin-binding subunit troponin T (TnT), and the inhibitory subunit troponin I (TnI). Troponin I binds to actin in thin myofilaments to hold the actin-tropomyosin complex in place, because of which myosin cannot bind to actin in relaxed muscle. When Calcium binds to the Troponin C it causes certain confomational changes which leads to dislocation of troponin I. The letter “I” is given due to its inhibitory character. | | | Catalog # | T8665-18P | | Applications | Suitable for use in ELISA and Western Blot. Other applications not tested. | | Recommended Dilution | Western Blot: 1:1000 | | Optimal dilutions to be determined by the researcher. | | Storage and Stability | May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. | | CAS Number | n/a | | Clone Type | Polyclonal | | Isotype | IgG | | Host | Rabbit | | Source | Human | | Concentration | As reported | | Form | Supplied as a liquid in 10mM sodium HEPES, pH 7.5, 150mM NaCl, 0.1mg/ml BSA, 50% glycerol. | | Purity | Purified by Protein A and immunoaffinity chromatography. | | Immunogen | Synthetic phosphopeptide corresponding to residues surrounding Ser23/24 of human troponin I (cardiac) (KLH). | | Specificity | Recognizes endogenous levels of human cardiac troponin I only when phosphorylated at serine 23/24 at 28kD.
