Mast cells contain a number of preformed chemical mediators such as histamine, chymase, carboxypeptidase and proteolytic tryptase. A substantial quantity of tryptase is estimated to be found in mast cells of skin and lung and suggestes this enzyme plays a major role in mast cell mediated events. In vitro studies indicate tryptase can cleave C3 to form C3a anaphylatoxin, inactivate fibrinogen as a coaguable substrate for thrombin and activate latent collagenase.

ApplicationsSuitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications not tested.
Recommended DilutionDirect ELISA: 0.5-1ug/ml. The detection limit for recombinant murine Tryptase beta1 is ~0.5ng/well.
Western Blot: 0.1-0.2ug/ml. The detection limit for recombinant murine Tryptase beta1 is ~1ng/lane under non-reducing and reducing conditions.
Immunoprecipitation: Immunoprecipitates recombinant, murine Tryptase beta1 from conditioned media of transfected NS0 cells. The recovered recombinant murine Tryptase beta1 can be detected by Western Blot analysis using a monoclonal antibody.
Optimal dilutions to be determined by the researcher.
Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

SpecificityRecognizes murine Tryptase, beta1 in direct ELISAs and Western Blots. Shows ~10% crossreactivity with recombinant human TPSB2, 5% crossreactivity with recombinant human TPS1 and less than 1% crossreactivity with recombinant human TPSG1 and recombinant murine MCPT1.
