| Tuberous sclerosis complex (TSC) is an autosomal dominant genetic disorder, which is characterized by seizures, mental retardation and hamartomatous in multiple organs. The genetic disorder is caused by the mutation in two genes - TSC1 and TSC2. The TSC2 gene is located on the chromosome 16p13.3, and it encodes the protein tuberin(1). Tuberin can interact with hamartin, encoded by TSC1 gene, to form a physical and functional complex(2). Tuberin, together with hamartin, are widely localized within epithelial cells, lymphocytes, endocrine glands and central nervous system (CNS) throughout the both normal body and TSC patients(1). Several independent investigations have demonstrated that tuberin is directly phosphorylated by Akt(2,3) and is further involved in stimulating cell growth, which provide an important link between tuberin and growth factor signaling.
