Embryonic vascular system undergoes a series of complex, highly regulated series of events involving differentiation, migration and association of primitive endothelial cells. This process is termed vasculogenesis. Study of tumor angiogenesis has led to the identification of several proteins including basic fibroblast growth factor (bFGF) and vascular endothelial growth factor. VEGF acts by interacting with a family of largely endothelial-specific receptor tyrosine kinases that includes VEGFR-1 (flt-1/flk-2), VEGFR-2 (flk-1/KDR), and VEGFR-3/Flt-4. Disruption of VEGFRs interferes with differentiation of endothelial cells and it is lethal for the embryo. FLT-1 (fms-like tyrosine kinase or VEGF-R1; human 1338 aa; rat 1336aa, and mouse 1333), a putative receptor protein tyrosine kinase, is a receptor for VEGF and PLGF. Mature full length protein is aa27-1338 (EC domain 27-758 aa). It is type 1 membrane protein and also exist as soluble protein. FLT-1 region aa657-687 (DQEAPYLLRNLSDHTVAISSSTTLD
CHANGVGEHCNK) is changed to KAVFSRISKFKSTRNDCTTQSNVKH in soluble FLT- 1. Additional isoforms seem to exist as well. Mostly in normal lung, but also in placenta, liver, kidney, heart and brain tissues. Specifically expressed in most of the vascular endothelial cells, and also expressed in peripheral blood monocytes. It is not expressed in tumor cell lines. Soluble Isoform (sFlt1) is strongly expressed in placenta.

Suitable for use in Western Blot, ELISA and Flow Cytometry. Other applications not tested.

Recommended DilutionsWestern Blot: 0.5-2ug/ml. Human recombinant baculovirus expressed VEGFR1 at ~166-200kD.
ELISA: 1:10,000-1:100,000
Flow Cytometry: 25ug/ml.10-20ul of diluted antibody labels 1-2x10e5 cells in a total volume of 200-250ul.
Optimal dilutions to be determined by the researcher.
Storage and StabilityLyophilized powder may be stored at -20°C. Stable for 12 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
