Vav proteins belong to the Dbl family of guanine nucleotide exchange factors (GEFs) for Rho/Rac small GTPases. Three mammalian members Vav1, Vav2 and Vav3, have been identified. Vav1 is expressed only in hematopoietic cells and is involved in the formation of the immune synapse. Vav2 and Vav3 are more ubiquitously expressed. In addition to the Dbl homology (DH) domain, which confers GEF activity, Vavs contain protein interaction domains that allow them to function in various pathways regulating organization of the actin cytoskeleton. Phosphorylation stimulates the GEF activity of Vav protein towards Rho/Rac.

Applications
Suitable for use in Western Blot and Flow Cytometry. Other applications not tested.
Recommended Dilutions
Western Blot: 1:1000. Incubate membrane with diluted antibody in 1X TBS, 5% BSA, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Flow Cytometry (Fixed/Permeabilized): 1:25 Optimal dilution determined by the researcher.
Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Immunogen
Synthetic peptide corresponding to residues surrounding Tyr174 of human Vav.
Form
Supplied as liquid in 10mM HEPES, pH 7.5, 150mM sodium chloride, 0.1mg/ml BSA, 50% glycerol. No preservative added.
Purity
Purified by Protein A and peptide affinity chromatography.
Specificity
Recognizes endogenous levels of total human Vav1 protein. Does not cross-react with Vav2, Vav3 or CelVav. Species Crossreactivity: mouse and rat
