Wnts have diverse roles in governing cell fate, proliferation, migration and polarity during development, particularly in embryogenesis, and also play a role in carcinogenesis in adults1-3. Wnts are transduced through at least three distinct intracellular signaling pathways, including the canonical Wnt/b-catenin, Wnt/Ca2+ and Wnt polarity (also called ‘planar polarity’)2, 4,5 pathways. Distinct sets of Wnt and Frizzled ligand-receptor pairs activate each of these pathways and lead to unique cellular responses. The Wnt/b-catenin pathway primarily regulates cell fate determination during development, whereas the main function of the Wnt polarity pathway is the regulation of cytoskeletal organization.

Chicken Wnt6 full-length cDNA was recently isolated6, and shares 73% and 74% identity with human and mouse Wnt6, respectively. Wnt6 is expressed in the ectoderm of chicken embryos adjacent to the neural plate and folds when neural crest cells are being induced, indicating that Wnt signaling may be a common mechanism for vertebrate crest induction.7 Wnt6 expression is observed from chicken embryo HH-stage 4 onwards and it can be seen in the ectoderm overlying the segmental and the epithelial somite at HH-stages 7-16.6 Further sites of expression are the boundary of the early neural plate and surface ectoderm, the roof of mesancephalon, pretectum and dorsal thalamus, the differentiating heart, and the otic vesicle.8 The precise sites of Wnt6 expression coincide with crucial changes in tissue architecture, namely epithelial remodeling and epithelial-mesenchymal transformation (EMT).8 Wnt6 expression domains are associated with sites of BMP signaling.8,9

Storage and StabilityMay be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

ImmunogenSynthetic peptide derived from an internal region of the chicken Wnt6 protein.
SpecificityReacts with the chicken Wnt6 protein. On Western blots, it identifies a band at ~35kD and one at ~45kD. The two bands may represent different isoforms or post-translational modification states. Immunohistochemical and immunofluoresent staining using this antibody is observed in non-neural ectoderm of chick embryos from stages 8 to 14, consistent with the reported expression of Wnt6 mRNA as determined by in situ hybridization. Additional staining is found in ectodermal tissues caudal to the otic level in heart- and gut-forming regions. Reactivity has been confirmed with stage 8-20 chicken embryo homogenates.
