| Mouse Wnt-8b is a 40-50kD secreted glycoprotein member of the Wnt family of proteins. Mature mouse Wnt-8b is 329aa in length and contains 24 cysteines plus three potential N-linked glycosylation sites. Little is known about Wnt8b. It appears to play a role in forebrain development. |

| Recommended Dilutions | Immunohistochemistry: 10ug/ml detects Wnt-8b on mouse fetal brain tissue sections. Sections were fixed with PBS containing 4% paraformaldehyde for 20 minutes at RT and blocked with PBS containing 10% normal donkey serum, 0.1% Triton X-100, and 1% BSA for 45 minutes at RT. After blocking, cells were incubated with diluted W3037-10 overnight at 4°C followed by Rhodamine Red-coupled anti-rat IgG at RT in the dark for one hour. Between each step, cells were washed with PBS containing 0.1% BSA. |
| Western Blot: 1-2ug/ml with the appropriate secondary reagents detects human or mouse Wnt-8b. Using a colorimetric detection system, the detection limit is ~25ng/lane under non-reducing and reducing conditions. Chemiluminescent detection will increase sensitivity by 5 to 50 fold. |
| Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~5ng/well. |

| Storage and Stability | Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute by adding sterile 40-50% glycerol, PBS. Reconstituted product is stable for 12 months at -20ºC. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
