Overdigestion AssayNo detectable change in the specific fragmentation pattern is observed after 320-fold overdigestion (20u/ug lambda DNA x 16 hours) with XagI.
Ligation/Recutting AssayAfter 50-fold overdigestion (3u/ug DNA x 17 hours) with XagI, approximately 80% of the DNA fragments can be ligated in the reaction mixture containing 2040u of T4 DNA Ligase/1ug of fragments and 10% of PEG at a 5'-termini concentration of 0.1µM. More than 95% of these can be recut.
Labeled Oligonucleotide (LO) AssayNo detectable degradation of a single-stranded and double-stranded labeled oligonucleotide was observed after incubation with 10 units of restriction endonuclease for 4 hours.
