Chemokine (C-X-C motif) ligand 3 (CXCL3) is a small cytokine belonging to the CXC chemokine family. It is mapped to 14p2 in rat. CXCL3 controls migration and adhesion of monocytes and mediates its effects on its target cell by interacting with a cell surface chemokine receptor. It has been shown that CXCL3 regulates cell autonomously the migration of the precursors of cerebellar granule neurons toward the internal layers of cerebellum, during the morphogenesis of cerebellum. CXCL3 also play fundamental roles in the development, homeostasis and it has effects on cells of the central nervous system as well as on endothelial cells involved in angiogenesis or angiostasis.
Sample Type
Cell culture supernates, serum and plasma (heparin, EDTA)
Intended Use
For quantitative detection of rat CXCL3 in cell culture supernates, serum and plasma (heparin, EDTA).
Specificity
Natural and recombinant rat CXCL3
Test Principle
This rat CXCL3 BioAssay™ ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for CXCL3 has been precoated onto 96-well plates. Standards(E.coli, R33-S101) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for CXCL3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the rat CXCL3 amount of sample captured in plate.
