Activin Receptor Type IIA (RIIA)
Members of the TGF-beta superfamily exert their biological effects by binding to heteromeric complexes of a type I and a type II serine-threonine kinase receptor, both of which are essential for signal transduction. Activin receptor type IIB is a type II receptor that mediates multiple signals for transforming growth factor-beta (TGF-beta) family members, including Activin, Nodal, Bmp7, Gdf1, Gdf3, Myostatin (Gdf8), and Gdf11. Through alternative mRNA splicing, multiple ActR-IIB isoforms can also be generated.

Suitable for use in Neutralization, ELISA, Western Blot and Immunohistochemistry. Other applications not tested.

Recommended Dilution:
Neutralization: ~1-3ug/ml will block 50% of the binding of 30ng/ml recombinant human Activin A to immobilized recombinant human Act RIIA/Fc Chimera (100ul of a 0.5ug/ml solution was coated in each well) in a functional ELISA assay.
Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~0.3ng/well.
Western Blot: 0.1-0.2ug/ml. The detection limit is ~5ng/lane and 25ng/lane under non-reducing and reducing conditions, respectively.
Immunohistochemistry (Paraffin): 15ug/ml for 5-15um thick sections. For detection of labeling, the ABC technique using chromogenic substrates is recommended.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
Lyophilized powder may be stored at -20C. Stable for 12 months at -20C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
