Description: ADAM19 (a disintegrin and metalloprotease 19; also MADDAM and Meltrin- ) is a 95-100kD member of the M12B peptidase family of enzymes. It is expressed on multiple cell types including monocytes, fibroblasts, osteoblasts and dendritic cells. The human ADAM19 proenzyme is a 931 amino acid (aa) type I transmembrane protein. It contains a 178 aa cleavable proregion (aa 26-203) and a 497 aa extracellular domain (ECD) (aa 204-702). The ECD contains a metalloprotease domain (aa 211-409), a disintegrin region (aa 417-503) and a Cys-rich domain (aa 504-650). The ECD circulates in plasma bound to 700kD 2-Macroglobulin. One splice variant shows an alternate start site at Met71, while two isoforms show variations afterLys902. Over aa 205-702, human ADAM19 is 87% aa identical to mouse ADAM19. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: ELISA: 0.5-1.0ug/ml Western Blot: 0.1-0.2ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
