Description: FYB (FYN binding protein) was initially identified as a molecular adaptor that couples T cell receptor (TCR) stimulation to the avidity of integrins governing T cell adhesion. TCR stimulation promotes the formation of a multi-protein complex containing CARMA1, MALT1, and BCL-10, which through the association of FYB, ultimately activates the NFkB family of transcription factors. More recent experiments have shown that FYB controls optimal T cell proliferation, cytokine production, and expression of the Bcl-2 family member Bclx(L), suggesting that FYB regulates T cell activation by promoting antigen-dependent T cell-antigen presenting cell (APC) activation. At least three isoforms of FYB are known to exist. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Western Blot: 0.5-2.0ug/ml detected ADAP/SLAP-130/Fyb in RIPA lysates from RAW 264.7 macrophage cells. RAW 264.7 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with A0859-43. Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system. Immunoprecipitation: 4ug immunoprecipitated ADAP/SLAP-130/Fyb from 500ug of RAW 264.7 macrophage RIPA lysate. Optimal dilutions to be determined by the researcher. Recommended Secondary Antibodies: I1904-39: IgG, X-Adsorbed (HRP) Pab Gt x Rb I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt x Rb I1904-40J: IgG, Fc (HRP) Pab Gt x Rb Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
