Description: This aggrecan degradation product usually remains within the tissue still complexed to hyaluronan and link protein. Its release from the cartilage usually signals that there has been extensive catabolism of aggrecan, which allows large complexes containing this metabolite to be released from the tissue. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Western Blot: 1:50 Immunohistochemistry: formalin or paraformalin paraffin embedded sections or alcohol-fixed frozen or unfixed snap-frozen sections. Notes: Samples must be deglycosylated using 0.01 Units Chondroitinase ABC, 0.01 Units Keratanase and 0.0001 Units Keratanase II per 10ug S-GAG of non-deglycosylated aggrecan for optimal epitope recognition. see Little et al. and Caterson et al. Diluted Technomouse culture supernatant. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
