Technical Data
Akt, phosphorylated (Ser473) (Rac PKa, PKBa)
Akt is a component of the PI-3 kinase pathway and is activated by phosphorylation at Ser473 and Thr308. Akt is a cytoplasmic protein also known as Protein Kinase B (PKB) and rac (related to A and C kinases). Akt is a key regulator of many signal transduction pathways. Akt exhibits tight control over cell proliferation and cell viability. Overexpression or inappropriate activation of Akt is noted in many types of cancer. Akt mediates many of the downstream events of PI 3-kinase (a lipid kinase activated by growth factors, cytokines and insulin). PI 3-kinase recruits Akt to the membrane, where it is activated by PDK1 phosphorylation. Once phosphorylated, Akt dissociates from the membrane and phosphorylates targets in the cytoplasm and the cell nucleus. Akt has two main roles: inhibition of apoptosis and promotion of proliferation.

Suitable for use in Immunofluorescence, ELISA, Western Blot and Immunohistochemistry. Other applications not tested.

Recommended Dilutions:
ELISA: 1:15,000-1:60,000
Western Blot: 1:200-1:1000, Detects a band at ~55.7kD
Immunohistochemistry (FFPE): 1:100-1:500. No pretreatment required.
Optimal dilutions to be determined by the researcher.

Recommended Secondary Antibodies:
I1904-39: IgG, X-Adsorbed (HRP) Pab Gt x Rb
I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt x Rb
I1904-46Q: IgG, H&L (HRP) Pab Gt x Rb

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
PabIgGAffinity Purified
100ug-20CBlue IceHumanRabbit
Synthetic peptide corresponding to the C-terminus of aa460-480 of human, mouse, rat and chicken Akt proteins, conjugated to KLH.
Purified by immunoaffinity chromatography
Supplied as liquid in PBS, pH 7.2, 0.01% sodium azide. No stabilizing proteins added.
Recognizes phosphorylated human Akt. Minimal reactivity occurs against non-phosphorylated Akt. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-rabbit serum. Species Crossreactivity: mouse and rat
Intended for research use only. Not for use in human, therapeutic, or diagnostic applications.
