Description: Directed against the region of AR which binds to heparin and is highly specific to AR. It shows no crossreaction with EGF, TGFa, or HB-EGF. The mature secreted form AR is an 84-amino acid residue glycosylated polypeptide growth regulator which is generated by proteolytic processing of a 252-amino acid transmembrane precursor. Seven different polypeptide ligands, which derive from distinct genes, are capable of binding to the extracellular domain of EGFR. These ligands include EGF, TGFa, AR, HB-EGF, cripto-1, epiregulin, and betacellulin. All of these growth factors contain a characteristic EGF-like domain which is defined by 6 evenly spaced cysteine residues that generate 3 loops through the formation of disulfide bonds. AR expression is induced by phorbol ester, estrogen, androgen, and other ligands of EGFR. In vitro, AR functions as an autocrine growth stimulator to drive proliferation of colon carcinoma cells, normal and oncogene-transformed mammary epithelial cells, cervical carcinoma cells, prostate cancer cells, and keratinocytes. AR is important in development of human placenta and murine mammary gland. AR is reportedly overexpressed in human cancers of breast, colon, stomach and pancreas. Applications: Suitable for use in Immunoprecipitation and Western Blot. Other applications have not been tested. Recommended Dilutions: Immunoprecipitation (Native verified): 10ug/mg protein lysate. Use Protein A. Western Blot: 5ug/ml for 2hrs at RT Optimal dilutions to be determined by the researcher. Positive Control: LS174T cells; HT29 cells Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
