Media used for enumeration of Aeromonas species in water by membrane filtration. Directions per Liter: Suspend 37.3 grams in liter of distilled or deionized water. Stir to mix thoroughly. Heat to boiling to dissolve completely. Autoclave for 15 minutes at 121ºC (15psi). Cool to 45-50°C. Add 10mg of sterile Ampicillin to inhibit E. faecalis. Alternatively, an additional 2mg sterile Vancomycin may be added to create Ampicillin Dextrin Agar with Vancomycin (ADA-V) to enumerate Aeromonas in finished waters. If detecting Aeromonas spp. in sea water, 50mg of the vibriostatic agent O129 can be added to the ADA medium. Storage and Stability: Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept refrigerated and used within a short period of time.
