Description: Angiopoietin-like 3 (ANGPTL3) is a secreted glycoprotein that belongs to the angiopoietin family. It is expressed in the liver and circulates as 70kD, 50kD, and 32kD species. These include full length ANGPTL3 as well as the proteolytically separated N- and C-terminal fragments which contain a coiled coil domain and fibrinogen-like domain, respectively. ANGPTL3 promotes an increase in circulating triglyceride levels by directly inhibiting lipoprotein lipase. Mature human ANGPTL3 shares 77% aa sequence identity with mouse ANGPTL3. Applications: Suitable for use in Western Blot and ELISA. Other applications not tested. Recommended Dilutions: Western Blot: 1-2ug/ml Using a colorimetric detection system, the detection limit for rhANGPTL3 is ~25ng/lane under non-reducing and reducing conditions. Chemiluminescent detection will increase sensitivity by 5 to 50 fold. ELISA (direct): 0.5-1.0ug/ml Detects human ANGPTL3. The detection limit for rhANGPTL3 is ~6.5ng/well. Optimal dilutions to be determined by the researcher. Hybridoma: Sf-21-derived myeloma cells with spleen B cells from Balb/c mice. Storage and Stability: Lyophilized powder may be stored at 4°C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol, PBS. Aliquot and store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
