Apolipoprotein AI (Apo AI)
Apolipoprotein AI promotes cholesterol efflux from tissues to the liver for excretion. Apolipoprotein AI is the major protein component of high density lipoprotein (HDL) in the plasma. Synthesized in the liver and small intestine, it consists of two identical chains of 77aa; an 18aa signal peptide is removed co-translationally and a 6aa propeptide is cleaved post-translationally. Apolipoprotein A I is a cofactor for lecithin cholesterolacyltransferase (LCAT) which is responsible for the formation of most plasma cholesteryl esters. Defects in the Apolipoprotein A I gene are associated with HDL deficiency and Tangier disease. The therapeutic potential of apoA-I has been recently assessed in patients with acute coronary syndromes, using a recombinant form of a naturally occurring variant of apoA-I. The availability of recombinant normal apoA-I should facilitate further investigation into the potential usefulness of apoA-I in preventing atherosclerotic vascular diseases.

Suitable for use in ELISA, Immunohistochemistry and Western blot. Other applications not tested.

Recommended Dilutions:
Western Blot: 1:250-1:500
ELISA: 1:2500-1:5000. Assayed against 1ug of Apolipoprotein in a standard ELISA using I1904-28K (IgG, H&L (HRP)) RbxGt and ABTS (A0030) for 30 minutes at RT.
Immunohistochemistry (Paraffin-embedded sections): 1:125-1:250
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Supplied as a liquid in 0.125M sodium borate, 0.075M sodium chloride, 0.005M EDTA, pH 8.0, 0.01% sodium azide, before the addition of glycerol to 40%.
Recognizes mouse Apolipoprotein AI (Apo AI). Minimum cross-reactivity to Apo AII, Apo B, Apo CI, Apo CII, Apo CIII, Apo E and Apo J. Non-specific crossreaction with other mouse serum proteins is negligible.
