Description: The Arp2/3 protein complex has been implicated in the control of actin polymerization in cells and has been conserved through evolution. The exact role of the protein encoded by this gene, the p34 subunit, has yet to be determined. Two alternatively spliced variants have been characterized to date. Additional alternatively spliced variants have been described but their full length nature has not been determined. Applications: Suitable for use in Western Blot, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5-2ug/ml detects p34-ARC in RIPA lysates from human A431, Jurkat and mouse 3T3 cells. A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-p34-Arc (1ug/ml). Proteins were visualized using a goat anti-rabbit IgG labeled with HRP* and a chemiluminescence detection system. Immunohistochemistry: 10ug/ml detects p34-Arc in mouse array tissue and formalin-fixed, paraffin-embedded human adenocarcinoma tissue. Immunocytochemistry: Detects p34-Arc/ARPC2 in C2C12 cells. Optimal dilution determined by the researcher. *Recommended Secondary Antibodies I1904-39: IgG, X-Adsorbed (HRP) Pab Gt xRb I1904-40A: IgG, H&L, X-Adsorbed (HRP) Pab Gt xRb I1904-46Q: IgG, H&L (HRP) Pab Gt xRb Included Positive Antigen Control: A0001-50: A431 Cell Lysate, Non-Stimulated. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
