Description: The INK4a-ARF locus encodes two unrelated proteins both of which function in tumor suppression. p14ARF arrests the cell cycle in a p53-dependent manner. p14ARF binds to mdm2 and promotes the rapid degradation of mdm2 protein required for mdm2 modification and concurrent p53 stabilization and accumulation. This interaction is mediated by the exon 1b-encoded N-terminal domain of p14ARF and a C-terminal region of mdm2. Deletion of the ARF-INK4a locus simultaneously impairs both the INK4a-cyclin D/cdk4-RB and the ARF-mdm2-p53 pathways. Applications: Suitable for use in Immunofluorescence, Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilution: Western Blot: 5-10ug/ml for 2 hrs. at RT Immunohistochemistry (Formalin/paraffin): 5-10ug/ml for 30 min. at RT. Note: Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min. followed by cooling at RT for 20 min. Optimal dilutions to be determined by the researcher. Positive Control: HeLa cells, cervical carcinoma or breast fibroadenoma. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
