Description: Applications: Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications not tested. Recommended Dilutions: Western Blot: A 1:500-1:1000 detects methylated p60 (unidentified protein) and Sm protein B, B’ and D in RIPA lysates from Jurkat cells. Jurkat cell lysate was resolved by electrophoresis, transferred to PVDF and probed with A3550-07C (1:1000). Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system. Detects bands at 60, 30, 32 and 16kD, corresponding to p60, Sm B, B’ and D, respectively. Immunoprecipitation: Reported by an independent laboratory to immunoprecipitate proteins containing symmetric dimethyl-arginine. Immunocytochemistry: 1:500. Reported by an independent laboratory to immunostain Cajal bodies in the nuclei of HeLa, WI-38 and NIH/3T3 cells fixed with paraformaldehyde and permeabilized with triton. Optimal dilutions to be determined by the researcher. Included Positive Antigen Control: J9000: Jurkat lysate. Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
