Description: Axin-1 is constitutively expressed and acts as a negative regulator of Wnt expression. It has been demonstrated that mouse Wnt, Wnt3a, is secreted by multiple cell lines and can act and as a growth factor for stem cells. Secreted Wnt3a is highly hydrophobic because it is palmitoylated on an essential cysteine C77; the palmitoylated state of Wnt3a is essential for stem cell signaling. Incubation of stem cells with Wnt3a in tissue culture medium resulted in the increased number of mouse hematopoietic stem cells expressing "primitive" phenotype. Applications: Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: ELISA: 1:128,000 Western Blot: 0.1ug/ml Immunohistochemistry (Paraffin): 3-5ug/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
