Cadherin 11 (OB-Cadherin, Osteoblast, CDH11)
Description:
Cadherins are a large family of Ca2+-dependent cell-cell adhesion molecules. All cadherins are single-pass transmembrane proteins with a variable number of 110 amino acid extracellular cadherin (EC) domains. Classical cadherins contain five EC domains and a conserved cytoplasmic domain. Based on amino acid alignment, classical cadherins are divided into type I and type II subgroups. Type I cadherins, including cadherins E (epithelial), N (neural), P (placental), and R (retinal) cadherin differ from type I cadherins in their specific amino acid sequences. Type II cadherins include human cadherin-5, -6, -8, -11, and –12. Cadherin 11 (OB-Cadherin) is a marker of the loosely connected and migratory cellular elements of the mesenchyme. Strong expression of Cadherin 11 has been noted in brain, spinal cord, bone marrow and bone cells, and is regulated in human endometrial glandular epithelial and stromal cells. A variant cadherin-11 gene (variant form) encodes a truncated protein with an unusual cytoplasmic domain not present in other cadherins. The variant protein is generated by alternative splicing, utilizing a 179 bp intronic sequence. Recently, Cadherin-11 mRNA and protein were found to be expressed in most invasive breast cancer cell lines tested but not in non-invasive cell lines, suggesting that cadherin-11 may be a marker for aggressive, invasive tumor subsets.

Applications:
Suitable for use in ELISA, Western Blot and Immunohistochemistry. Other applications not tested.
Recommended Dilutions:
ELISA: 0.1-1ug/ml
Western Blot: 1-2ug/ml
Immunohistochemistry (Frozen, FFPE): 1-3ug/ml. Heat induced epitope retrieval in citrate buffer, pH 6.0, is required. To stain, dilute 1:50-1:100. Incubate for 30-60 minutes at RT or overnight at 4°C.
Optimal dilutions to be determined by the researcher.
Western Blot Positive Control:
MDA.MB231 cells (human) and mouse L broblast lysates

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Specificity:
Recognizes human Cadherin 11 at ~110kD. An additional band at ~130kD (presumably the Cadherin 11 precursor) may also be observed. Some crossreactivity with other Cadherins is observed in approximately the following order: Cadherin11>>R-Cadherin>Cadherin 8>>N-Cadherin=P-Cadherin. No crossreactivity with Cadherin 6 or E-Cadherin has been observed. Species Crossreactivity: mouse
