Description:
Applications:
Suitable for use in Kinase assay and Western Blot. Other applications have not been tested.

Recommended Dilution:
Kinase Assay: 0.5-2ug/ml. Active MAP Kinase 2/Erk2 (Catalog #M2362-06) was used to phosphorylate serine 789 of purified porcine (hog) stomach Caldesmon.
Western Blot: 2ug/ml. Detects phosphorlyated isoform of Caldesmon in a RIPA lysate of EGF stimulated human A431 cells. Pretreating blots with 40U/ml of lambda phosphatase prior to incubation abolished the detection of Caldesmon.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Supplied as a liquid in 0.1M Tris-glycine, pH 7.4, 0.15M sodium chloride, 0.05% sodium azide, before the addition of glycerol to 30%.
Specificity:
Recognizes human Caldesmon phosphorylated at Ser789 at ~Mr 140kD along with several lower molecular weight isoforms. Species Crossreactivity: porcine, canine and ferret. Species sequence homology: rat
