Caveolae are membrane invaginations that are prominent on the surface of endothelial cells, smooth muscle cells and fibroblasts. Numerous morphological studies indicate that caveolae function in the transcytosis of certain blood-borne macromolecules. Caveolin is a 21-24kD protein which was originally identified as a major v-Src substrate in RSV-transformed cells. Recently, caveolin has been identified as an integral membrane protein which is an important structural and regulatory component of the caveolae membranes. Two major isoforms of caveolin are known to exist: a 21kD species (beta-isoform) and a 24kD species (alpha-isoform). Both contain a complete C-terminus but differ in the N-terminal sequence.

Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications have not been tested.

Recommended Dilution:
Western Blot: 0.5-2ug/ml. Detects caveolin in RIPA lysates from EGF stimulated A431 cells. Since caveolin is an integral membrane protein, the use of membrane preparations may enhance detection. EGF stimulated A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Human caveolin (1ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
Immunoprecipitation: Reported to immunoprecipitate from human, monkey and rat fibroblasts.
Immunocytochemistry: Caveolin staining has been reported in human fibroblasts.
Optimal dilutions to be determined by the researcher.

Positive Antigen Control:
EGF stimulated A431cell lysate. Use 20ug per lane for minigels.

Storage and Stability:
May be stored at 4C for short-term only. For long-term storage, store at -20C. Aliquots are stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
