Chemokines are a superfamily of small proteins involved both in routine leukocyte trafficking and in the activation and recruitment of specific cell populations to sites of inflammation an infection. The biological activity of chemokines is mediated by their interaction with a family of seven-transmembrane G protein-coupled receptors, including CCR1, CCR2A, CCR3, CCR4, CCR5, as well as the Duffy blood group antigen and several virally encoded chemokine receptors (1). The C-C receptors are important in the function of T cell chemotaxis and migration of phagocytic cells to sites of inflammation. Specific chemokine receptor-ligand pairs are associated with particular disease states or tissue-specific lymphocyte homing, for example MDC-CCR4 plays an important role in the formation of T cell-dendritic cell clusters both in inflamed skin and
lymph nodes (2). CCR4 3-4 is a high affinity receptor for two recently described CC chemokines, thymus and activation-regulated chemokine (TARC)5 and macrophage-derived chemokine (MDC) (5). CCR4 mRNA is expressed predominantly in the thymus, spleen, and in peripheral blood leukocytes, including T cells, basophils, monocytes4 , macrophages, and platelets6. CCR4 is also expressed on all skin homing T cells expressing cutaneous lymphocyte antigen (CLA)(7). CCR4 has a key role in the inflammatory response leading to LPS-induced toxic shock (8).

Suitable for use in Western Blot. Other applications not tested.

Recommended Dilutions:
Western Blot: 1-3ug/ml
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes mouse CCR 4 at ~45 kD. Species Crossreactivity: rat. Reactivity has been confirmed with mouse EL-4 and SVEC4-10 cell lysates, immature mouse thymus homogenates and rat thymus homogenates.
