CD31 (PECAM-1, Platelet gpIIa Molecule) (Biotin)
Description:
CD31 (PECAM-1, Platelet gpIIa Molecule), a ~130kD integral membrane glycoprotein, is expressed in platelets, cultured endothelial cells, monocytes, normal tissue macrophages in tonsil, lung and kidney, skin biopsies from conditions such as sarcoidosis and lepromatous leprosy and bone marrow-derived hematopoietic stem cells and embryonic stem cells. CD31 mediates cell-cell adhesion. Valuable in the study of benign and malignanat vascular tumors. Staining for CD31 has also been used to measure angiogenesis, which reportedly predicts tumor recurrence.

Applications:
Suitable for in Flow Cytometry, Immunoprecipitation, ELISA, Immunofluoresence and Immunohistochemistry. Other applications not tested.

Recommended Dilution:
Flow Cytometry: 1:50 stains the extracellular domain of mouse CD31. 10e6 cells were used per sample. No fixation or permeabilization needed. Epitope is sensitive to trypsin.
Immunoprecipitation: Validated using the lysate of 35S-Methionine labeled 5HV cells.
Immunohistochemistry (Frozen): 1:50, Tissue sections were fixed in acetone for 10 min at RT and pretreated with 0.01% hydrogen peroxide to quench endogenous peroxidases. Normal rabbit serum was used as a blocking agent.
Optimal dilutions to be determined by the researcher.

Positive Control:
Mouse t-end.1 cells

Negative Control:
L929 cells (Fibroblasts)

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Specificity:
Recognizes mouse form of the Platelet-Endothelial Cell Adhesion Molecule. Reactivity of the antibody is restricted to the isoform of the molecule that is selectively expressed by endothelial cells. The antibody precipitates a 130kD molecule present on the membrane of endothelial cell presents on all mouse blood vessels both in normal and inflamed or tumor tissues.
