CD36 (gpIIIb, gpIV, Thrombospondin Receptor)
CD36 (88kD), a cell surface class B scavenger receptor, is a heavily N-glycosylated transmembrane protein of ~88kD with two short intracellular domains and a large extracellular domain. CD36 is expressed on platelets, mature monocytes and macrophages, microvascular endothelial cells, mammary endothelial cells, during stages of erythroid cell development and on some macrophage derived dendritic cells.

Suitable for use in Immunoprecipitation, ELISA, Flow Cytometry, Immunofluorescence, Immunohistochemistry, Neutralization and Functional Studies. Other applications have not been tested.

Recommended Dilutions:
Immunoprecipitation: 10ug/200ug protein. Detected 88kD sialoglycoprotein in platelets; 85 and 88kD in HEL cells.
ELISA: 10ug/ml using 100ul/well as coating antibody diluted in Tris-buffered saline. Incubated overnight at RT (3).
Flow Cytometry: 2ug stains 10e5 cells. Stains the extracellular domain of CD36 in unfixed cells. Positive on granulocytes (1).
Immunofluorescence: Unfixed cells were incubated for 30 minutes at 4C followed by a secondary FITC Pab. Fixed with methanol for 1 minute before analysis (1).
Immunohistochemistry (Frozen): 1:50. Tissue embedded in tissue-tek (for instance aortic tissue) followed by freezing in liquid nitrogen. 7-8um sections were air-dried, fixed with acetone and blocked with 10% NGS (4).
Functional Studies: 1ug/ml induced platelet aggregation and secretion (2).
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Recognizes human CD36 (88kD). Sensitive for neuroaminidase, resulting in a shift from 88 to 85kD. Recognizes adult and fetal monocytes, platelets and reticulocytes. Does not stain lymphocytes or granulocytes. Reactivity has also been found in small intestine, kidney, liver and thyroid. Blocks the biological activity of CD36 by blocking collagen/thrombospondin binding. Agglutinates fetal but not adult erythrocytes.
