Description: Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. The I-domain containing alpha integrin 2 combines with beta integrin 1 to form a collagen-binding integrin referred to as glycoprotein Ia/IIa (gpIa/IIa) when expressed on platelets, and very late (activation) antigen 2 ('VLA-2') when found on T-cells. In addition to adhesion, integrins are known to participate in cell-surface mediated signalling. CD49b is expressed approximately 50% of monocytes and most adherent cell lines. CD49b forms a non-covalent association with the 130kD VLA beta chain (CD29) in the VLA-2 complex. The CD49b/CD29 complex is also known as a2/b1 and VLA-2. The complex mediates binding of cells to collagen (types I, II, III, and IV) and to laminin. The complex of CD49b associated with CD29 (integrin b1) is a receptor for laminin and collagen. This complex is expressed by some CD4-positive T-cells, natural killer cells, epithelial cells and platelets. Applications: Suitable for use in Immunohistochemistry. Other applications not tested. Recommended Dilution: Attachment inhibition assays: Fibroblasts, epithelial cells, endothelial cells, and non-activated platelets on collagen types I, III, IV, VI and laminin. Immunohistochemistry (immunofluorescence): Fresh frozen or paraffin embedded tissue (protease digestion is necessary when using paraffin). For extensive dilution, protein-containing or other stabilizing medium should be used. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
