CDC25C (Cell Division Cycle 25 Homolog C, Cdc 25C, CDC 25, Dual Specificity Phosphatase Cdc25C, M-phase Inducer Phosphatase 3, MPIP3, PPP1R60)
cdc25C (Mr 56 and 59kD), in asynchronous cells, is a dual specificity protein phosphatase that controls entry of cells into mitosis by dephosphorylating and activating the Cdc2/cyclin B complex. cdc25C is regulated at both the transcriptional and post-translational levels. Expression of cdc25C is restricted to late S/G2, effected by a transcriptional repressor that binds the CDE (cell cycle-dependent element) in the basal promoter. cdc25C is also regulated via phosphorylation of Ser216 by C-TAK1 (cdc25c associated protein kinase) which mediates interaction with the 14-3-3 family, maintaining cdc25C “inactive”. Phosphorylated Ser216 is detectable only throughout interphase. Cells expressing cdc25C mutants that cannot be phosphorylated at Ser216 exhibit perturbed mitotic timing and escape the G2 checkpoint arrest induced by radiation induce DNA damage.

Suitable for use in Immunoblot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.

Recommended Dilution:
Immunoblot: 1-4ug/ml detects cdc25C in HeLa nuclear extract. HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with C2560-05
(1ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody labeled with HRP and a chemiluminescence detection system.
Immunoprecipitation: Reported to immunoprecipitate cdc25C.
Immunohistochemistry: Reported to immunostain cdc25C.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
