Description: Cyclin-dependent kinases (cdk) are the catalytic subunits of the cyclin/cdk complexes, which phosphorylate substrates on threonine/serine residues. Cdk2 interacts with cyclin D1, D3 and E, as well as cyclin A. Cyclin E/cdk2 kinase is active in G1- and S-phase of the cell cycle and is important for progression of cells from G1 to S. Cyclin A/cdk2 levels are maximal at the S/G2-phase transition and both are found associated with DNA in the initiation complex during replication. Rb is a substrate for cyclin E/cdk2 and cyclin A/cdk2 in vivo. Applications: Suitable for use in Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: Immunoprecipitation: 10ul/mg; co-precipitates associated proteins Western Blot: 1ug/ml Incubate fot 2 hours at RT. Immunohistochemistry: 2-4ug/ml cryostat sections and paraffin embedded sections. For paraffin embedded sections, boil the sections in 1mM EDTA, pH 8, for 10-20 min followed by cooling at RT for 20 min. Optimal dilutions to be determined by the researcher. Hybridoma: P3-NS1/AG4-1 (NS1) myeloma cells with spleen cells from Balb/c mice. Positive Control: HeLa, 293, VA13, IMR90, Jurkat and WI-38 cells; thymus, tonsil or reactive lymph node. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
