Description: PSD-93 (Post Synaptic Density 93) belongs to a class of proteins referred to as MAGUKs (membrane-associated guanylate kinases). MAGUKs are widely expressed in the brain and are critical elements of the cytoskeleton and of certain synapses. Cannonical MAGUK proteins contain one or three PDZ (PSD-95/discs large/zona occludens-1) domains, an SH3 domain, and a guanylate kinase homology domain. These proteins appear to regulate synaptic function by acting as molecular scaffolds to organize and cluster signaling machinery at synapses, in part by interaction of their PDZ domains with carboxy-terminal T/SXV motifs present on synaptic proteins including NMDA receptors and Shaker type K+ channels. PSD-93 and several other MAGUK and PDZ containing proteins (eg. PSD-95, SAP-97, SAP-102, MALS/Veli proteins,) are enriched in the post synaptic density. PSD-93 is expressed in discrete neuronal populations as well as in specific nonneuronal cells and exhibits complex molecular diversity attributable to tissue-specific alternative splicing. PSD-93, like PSD-95, binds to nNOS and to the NMDA receptor 2B. PSD-93, however, is unique among PSD-95/SAP-90 family members in its expression in cerebellar Purkinje neuron cell bodies and dendrites. Applications: Suitable for use in Immunofluorescence, Immunoprecipitation and Western Blot. Other applications have not been tested. Recommended Dilution: Immunofluorescence: 1-3ug/ml Immunoprecipitation: 5-10ug/IP reaction Western Blot: 0.1-1ug/ml Optimal dilutions to be determined by the researcher. Positive control: rat brain. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
