Claudin 10 (CLDN10)
Tight junctions are intercellular junctions that act as both the barrier (or gate), and the fence. The barrier function of tight junctions regulates the passage of ions, water, and various macromolecules through paracellular spaces. The fence function maintains cell polarity, thus, tight junctions work as a fence to prevent intermixing of molecules in the apical membrane with those in the lateral membrane. Tight junctions are comprised of the membrane proteins occludin, claudins, and JAMs. Claudins are responsible for the formation of tight junction strands and are connected with the actin cytoskeleton mediated by ZO-1. Claudin 10 is concentrated at the tight junctions of epithelial cells of AQP1-positive tubules in the kidney cortex (proximal tubules). Claudin 10 is also detected at the tight junctions of epithelial cells of THP-positive tubules in the kidney medulla (thick ascending limb of Henle).

Suitable for use in Western Blot, Immunohistochemistry and Immunofluorescence. Other applications have not been tested.

Recommended Dilution:
Western Blot (Mouse, rat): 1-3ug/ml. Identifies a band at ~21kD. Bands of unknown origin are observed at ~75kD, ~70kD, ~35kD and/or ~17kD in some homogenates. When using mouse brain homogenates a single band at ~21kD was observed.
Immunohistochemistry (Mouse) (Frozen): 3-6ug/ml
Immunofluorescence (Mouse): 3-6ug/ml
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
