CNPase (2',3'-cyclic Nucleotide 3'-phosphodiesterase)
Description:
CNPase (2,3-Cyclic Nucleotide 3-Phosphodiesterase) exists in two forms: 46kD (CNP1) and 48kD (CNP2). It is expressed at high levels by oligodendrocytes in the central nervous system and by Schwann cells in the peripheral nervous system and is virtually absent in other cell types. CNPase activity is decreased in demyelinating diseases such as multiple sclerosis. CNPase gene is localized to chromosome 17q21, very near the BRCA1 familial breast cancer gene.

Applications:
Suitable for use in ELISA, Immunofluorescence, Western Blot, and Immunohistochemistry. Other applications have not been tested.

Recommended Dilution:
Immunohistochemistry (formalin/paraffin): 2-4ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 minutes followed by cooling at RT for 20 minutes.
Optimal dilutions to be determined by the researcher.

Positive Control:
Brain

Storage and Stability:
May be stored at 4C for short-term only. For long-term storage, aliquot and store at -20C. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Immunogen:
Purified human 2'-3'-cyclic nucleotide 3'-phosphodiesterase (CNPase). Cellular Localization: Cell membrane.
Purity:
Purified by Protein G affinity chromatography.
Form
Supplied as a liquid in PBS, pH 7.4, 0.2% BSA, 0.1% sodium azide, before the addition of glyceol to 40%. Also available without BSA and azide. See C6200X.
Specificity:
Recognizes human proteins of 46kD (CNP1) and 48kD (CNP2), identified as two forms of CNPase. This clone can be used for early identification of oligodendrocytes and Schwann cells. Species Crossreactivity: Human, monkey, bovine, sheep, goat, porcine, feline, canine, rabbit, rat and mouse. Does not react with guinea pig or chicken.
Intended for research use only. Not for use in human, therapeutic, or diagnostic applications.
