Description: Applications: Suitable for use in Western Blot, Immunoprecipitation, Immunocytochemistry and Supershift Assay. Other applications have not been tested. Recommended Dilution: Western Blot Analysis: 0.1-2.0ug/ml detects CUG-BP1 in HeLa nuclear extract. HeLa nuclear extract was resolved by electrophoresis, transferred to nitrocellulose and probed with C7948-50 (0.5ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: Immunoprecipitates CUG-BP1 from HeLa cell lysates Immunocytochemistry: Nuclear staining in HeLa and myoblasts from normal patients. Supershift: Supershifts CUG-BP1:DNA complexes in EMSA Optimal dilutions to be determined by the researcher. Recommended secondary antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-14C: IgG, Fc (HRP) Pab Gt xMo I1904-19C: IgG, F(ab’)2 (HRP) Pab Gt xMo Included Positive Antigen Control: H1838: HeLa nuclear extract. Add an equal volume of Laemmli reducing sample buffer to 5-10ul of extract and boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for minigels. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
