DARPP32, NT (Dopamine and cAMP-regulated Phosphoprotein, Protein phosphatase 1 regulatory subunit 1B, PP1R1B)
The 32kD dopamine and cyclic adenosine 3',5'-monophosphate-regulated phosphoprotein (DARPP-32) can either function as a serine/threonine kinase or as a serine/threonine phosphatase depending upon which particular amino acid residue is phosphorylated(1). When phosphorylated by protein kinase A (PKA) at threonine 34, DARPP-32 becomes a potent inhibitor of protein phosphatase-1(2,3). Alternatively, when phosphorylated at threonine 75 by cyclin-dependent kinase 5 (Cdk5), DARPP-32 is converted into an inhibitor of PKA(1). DARPP-32 and truncated DARPP (t-DARPP) are overexpressed in gastric cancers(4).

Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry (frozen and paraffin), Immunocytochemistry, Immunofluorescence and Flow Cytometry. Other applications not tested.

Recommended Dilution:
Western Blot: 1:5000-1:50,000. Detects a band of ~32kD.
Immunoprecipitation: 1:50
Immunohistochemistry (frozen); 1:7,500
Immunocytochemistry: 1:50-1:100
Optimal dilutions to be determined by the researcher.

Immunohistochemistry: Human breast adenocarcinoma (Paraffin-embedded)

Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
