Description: DMP-1 (dentin matrix protein 1) is an acidic, 100-160kD member of the phosphophoryn family of extracellular matrix proteins. Mouse DMP-1 is found both intra- and extracellularly, where phosphorylated DMP-1 participates in bone mineralization. Secreted mature mouse DMP-1 is a 487 amino acid (aa) proteoglycan. It contains multiple serine, glutamic acid, and aspartic acid residues, a GAG attachment site (aa71-72) and an RGD motif (aa350-352). Multiple cleavage sites exist between aa205-253, creating multiple fragments ~37kD and 57kD in size. Applications: Suitable for use in Western Blot, Immunohistochemistry and ELISA. Other applications have not been tested. Recommended Dilutions: Western Blot: 0.1-0.2ug/ml The detection limit is ~1ng/lane and 5ng/lane under non-reducing and reducing conditions, respectively. Immunohistochemistry: 10ug/ml detects DMP1 in E15.5 mouse embryos. Fix cells with PBS, 4% paraformaldehyde for 20 minutes at RT and block with PBS, 10% normal donkey serum, 0.1% Triton X-100, 1% BSA for 45 minutes at RT. Incubate cells with D8075-51 for 3 hours at RT followed by donkey anti-sheep lgG at RT for 1 hour. Between each step, wash cells with PBS, 0.1% BSA. Direct ELISA: 0.5-1.0ug/ml The detection limit is ~0.1ng/well. Optimal dilutions to be determined by the researcher. Hybridoma: NSO myeloma cells with spleen cells from Balb/c mice. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. |
