Dulbecco’s MEM is the most widely used modification of BME. It contains a 4-fold higher concentration of amino acids and vitamins. Non-essential amino acids and certain essential trace elements were added. The bicarbonate concentration was increased. The standard formula for DMEM is with 1000mg/ml glucose. DMEM was originally developed for the culture of mice embryonic cells. Today, it finds a broad application of serum free culture of normal and transformed mouse and chicken cells. Directions per Liter Dissolve 12.9g in 800-900ml of ddH2O, stirring gently until completely solubilized. If required, add 3.7g sodium bicarbonate with stirring. Adjust pH of the medium to 0.1-0.3 pH unit below the desired level. Add additional water to bring the solution to 1L. Filter-sterilize using a 0.22 micron membrane filter. Aliquot into sterile containers. Do not autoclave. Contains heat-labile compounds that can be damaged with autoclaving. Storage Store powdered media at RT. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4°C and used within a short period of time.
