Estrogen Receptor (ER)
Description:
ER gene consists of more than 140kb of genomic DNA divided into 8 exons, being translated into a protein with six functionally discrete domains required for transcription activation function, binding to estrogen response element (ERE) constitutive dimerization, binding to heat shock proteins, and ligand recognition. The ER is an important regulator of growth and differentiation in the mammary gland. Presence of ER in breast tumors indicates an increased likelihood of response to anti-estrogen (e.g. tamoxifen) therapy.

Applications:
Suitable for use in Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications not tested.

Recommended Dilutions:
Immunoprecipitation (denatured verified): 10ug/mg protein lysate. Use Protein A.
Western Blot: 2.5-5ug/ml for 2hrs at RT.
Immunohistochemistry: 10-20ug/ml for 30 min at RT. Staining of formalin/paraffin tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min.
Optimal dilutions to be determined by the researcher.

Positive Control:
MCF-7 cells. Breast carcinoma

Storage and Stability:
May be stored at 4C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20C or colder. Aliquots are stable for at least 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
