FAK (Focal Adhesion Associated Protein Tyrosine Kinase, BC3)
FAK is a widely expressed non-receptor protein tyrosine kinase (PTK). FAK has a central catalytic domain and a C-terminal tail that localizes it to focal adhesions, which are sites where cells attach to the extracellular matrix via surface integrin receptors. Increased FAK tyrosine phosphorylation occurs upon integrin engagement with fibronectin. Studies have shown that adhesion of murine NIH3T3 fibroblasts to fibronectin promotes association of the Grb2 adapter protein and c-Src PTK with FAK in vivo, and also results in activation of the ERK2 MAP kinase. In v-Src-transformed NIH3T3, the association of v-Src, Grb2, and Sos with FAK is independent of cell adhesion to fibronectin. In vitro the Grb2 SH2 domain binds directly to tyrosine-phosphorylated FAK, and the binding site has been identified as Tyr925 by site directed mutagenesis.

Suitable for use in Immunofluorescence and Immunoprecipitation. Not suitable for Western Blot. Other applications not tested.

Recommended Dilution:
Immunofluorescence: 10ug/ml detects FAK in chicken embryo fibroblasts.
Immunoprecipitation: 4ug immunoprecipitates Fak from a 3T3/A31 cell lysate and previous lots from a Src-transformed chicken cell line lysate (500ug-1mg). Confirmed by subsequent immunoblot analysis of the immunoprecipitate using 1-2ug/ml of anti-FAK, clone 4B7 (F0019-55A).
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20C. Aliquots are stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
