Fibronectin (Biotin)
Description:
Fibronectins are disulfide-linked, dimeric glycoproteins of ~440kD. They possess at least four binding sites for collagen, glycosaminoglycans, transglutaminase, and a cell surface receptor. Fibronectins are involved in cell adhesion, tissue organization and wound healing. Fibronectins are present in basement membranes, interstitial connective tissue matrix and blood. Cellular fibronectin is widely distributed in the stroma of many malignant tumors.

Applications:
Suitable for use in ELISA, Immunoprecipitaion, Immunohistochemistry and Western Blot. Other applications not tested.

Recommended Dilutions:
ELISA: 1:4000-1:20,000. Assayed against 1ug of Fibronectin in a standard capture ELISA using ABTS (A0030) as a substrate for 30 minutes at RT.
Immunohistochemistry (Paraffin embedded tissue): 1:50-1:200
Western Blot: 1:500-1:10,000
Immunoprecipitation: 1:100
Optimal dilutions to be determined by the researcher.

Biotin/Protein Ratio:
10-20 BAC molecules per rabbit IgG molecule.

Storage and Stability:
Lyophilized powder may be stored at -20C. Stable for 12 months after receipt at -20C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20C. Reconstituted product is stable for 12 months at -20C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Supplied as a lyophilized powder from PBS, pH 7.2, 10mg/ml BSA (IgG and Protease free), 0.01% sodium azide. Labeled with Biotin. Reconstitute with 100ul sterile ddH2O.
Specificity:
Recognizes human Fibronectin. Species Crossreactivity: mouse, rat, bovine and monkey. Typically less than 1% crossreactivity against other extracellular matrix proteins was detected by ELISA against purified standards. Has negligible crossreactivity with Type I, II, III, IV, V or VI Collagens or Laminin. Non-specific crossreactivity with other human serum proteins or non-Fibronectin extracellular matrix proteins is negligible.
