This factor was found initially in conditioned medium of the rat B49 glial cell line. GDNF is a disulfide-bonded homodimeric glycosylated protein of 134 amino acids. The proteins contain seven conserved cysteine residues in the same relative spacing found in all members of the TGF-beta superfamily of proteins. The human GDNF gene has been mapped to chromosome 5p13.1-p13.3. Related factors are Neurturin, Persephin, and Artemin. Collectively they are being referred to as GDNF family ligands. The ED50, calculated by the dose-dependent dopamin uptake in rat mesencephalic cultures was found to be 5-10ng/ml. Storage and Stability: Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O, aliquot and store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
